The Bioaim Mouse aFGF ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of aFGF in mouse serum, plasma and cell culture media. An antibody specific for mouse aFGF was coated on a 96-well plate. Standards and samples are added to the wells and any aFGF present binds to the immobilized antibody. The wells are washed and biotinylated anti-mouse aFGF antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of aFGF present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.Research Area Angiogenesis; cell growth; Cancer Biomarker;
fibroblast growth factor 1 (aFGF) Background: FGF acidic, also known as FGF-1, ECGF, and HBGF-1, is a secreted mitogen that stimulates the proliferation of all cells of mesodermal origin and many cells of neuroectodermal, ectodermal, and endodermal origin. It plays a number of roles in development, regeneration, and angiogenesis. FGF acidic is released extracellularly as a disulfide-linked homodimer and is stored in complex with extracellular heparan sulfate. The association of FGF acidic with heparan sulfate is a prerequisite for its subsequent interaction with FGF receptors. Internalized FGF acidic can translocate to the cytosol and to the nucleus. Intracellular FGF acidic functions as a survival factor by inhibiting p53 activity and proapoptotic signaling. The protein encoded by this gene is a member of the fibroblast growth factor (FGF) family. This protein functions as a modifier of endothelial cell migration and proliferation, as well as an angiogenic factor. It acts as a mitogen for a variety of mesoderm- and neuroectoderm-derived cells in vitro, thus is thought to be involved in organogenesis. Three alternatively spliced variants encoding different isoforms have been described.
