The Bioaim Mouse ACE ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of ACE in mouse serum, plasma and cell culture media. An antibody specific for mouse ACE was coated on a 96-well plate. Standards and samples are added to the wells and any ACE present binds to the immobilized antibody. The wells are washed and biotinylated anti-mouse ACE antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of ACE present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: ACE and ACE-2, two cell surface proteases, are important regulators of the renin-angiotensin system (RAS), which plays a key role in maintaining blood pressure homeostasis and fluid salt balance in mammals. Because of its location and specificity, ACE plays additional roles in immunity, reproduction and neuropeptide regulation. ACE exists in two isoforms. Somatic ACE comprises two highly similar protease domains whereas germinal ACE comprises a single protease domain. ACE-2 consists of a single protease domain and is an essential regulator of heart and lung function. It is also a cellular receptor for the Spike protein of SARS cornonavirus that causes severe acute respiratory syndrome. ACE hydrolyzes peptides by the removal of a dipeptide from the C-terminus. Likewise it converts the inactive decapeptide angiotensin I to the octapeptide angiotensin II by removing the dipeptide His-Leu. Angiotensin II is potent vasoconstrictor in a substrate concentration-dependent manner. Angiotensin II binds to the type 1 angiotensin II receptor (AT1), which sets off a number of actions that result in vasoconstriction and therefore increased blood pressure.
