The Bioaim Human Uromodulin ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of Uromodulin in human serum, plasma and cell culture media. An antibody specific for human Uromodulin was coated on a 96- well plate. Standards and samples are added to the wells and any Uromodulin present binds to the immobilized antibody. The wells are washed and biotinylated anti-human Uromodulin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of Uromodulin present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.Research Area Kidney biomarker;
uromodulin (Uromodulin) Background: Uromodulin (also Tamm-Horsfall glycoprotein or THP) is an 85 – 95 kDa urinary glycoprotein. It is secreted by renal tubule epithelium, acts as a binding protein for IL-1, TNF-α and C1q, activates resting monocytes and promotes neutrophil phagocytosis. Uromodulin forms high molecular weight oligomers that line the kidney tubules. Human Uromodulin is GPI-linked. Uromodulin excretion in urine follows proteolytic cleavage of the ectodomain of its glycophosphatidylinositol-anchored counterpart that is situated on the luminal cell surface of the loop of Henle. Uromodulin may act as a constitutive inhibitor of calcium crystallization in renal fluids. The excretion of uromodulin in urine may provide defense against urinary tract infections caused by uropathogenic bacteria. Uropontin, nephrocalcin and uromodulin are the three known urinary glycoproteins that affect the formation of calcium-containing kidney stones or calculus. Tamm–Horsfall protein is part of the matrix in renal calculi but a role in kidney stone formation remains debatable. However, decreased levels of Tamm–Horsfall in urine have been found to be a good indicator of kidney stones.
