The Bioaim Human Syndecan 1 ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of Syndecan 1 in Human serum, plasma and cell culture media. An antibody specific for Human Syndecan 1 was coated on a 96-well plate. Standards and samples are added to the wells and any Syndecan 1 present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human Syndecan 1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of Syndecan 1 present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: Syndecans are type 1 transmembrane proteoglycans that constitute the major physiological form of heparan sulfate on the cell surface. The four vertebrate syndecans, Syndecan-1 through -4, have similar short cytoplasmic domains and extracellular portions that diverge, except for HS attachment sites. Structurally diverse side chains add considerably to the size of the core proteins and serve as binding sites for growth factors, cytokines, and extracellular matrix proteins. Syndecans are present as homodimers or multimers, and are often expressed in developmental and cell type-specific patterns.
