The Bioaim Human Osteoactivin ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of Osteoactivin in Human serum, plasma and cell culture media. An antibody specific for Human Osteoactivin was coated on a 96-well plate. Standards and samples are added to the wells and any Osteoactivin present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human Osteoactivin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of Osteoactivin present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.Research Area Inflammation;

glycoprotein nmb (Osteoactivin) Background: Osteoactivin, also known as transmembrane glycoprotein NMB (GPNMB), is a type I transmembrane glycoprotein that is found either on the plasma membrane or on membranes of the endosome/lysosome compartment. The extracellular region of Osteoactivin contains a heparin-binding motif and an integrin recognition RGD domain, while the cytocylasmic region contains an immunoreceptor tyrosine-based activation motif and lysosomal targeting motif. In osteoblast progenitor cells, Osteoactivin works as a positive regulator of osteoblast differentiation during later stages of matrix maturation and mineralization that is mediated at least in part by BMP-2 in a SMAD1 dependent manner to promote osteoblast differentiation.
