The Bioaim Human Kininogen ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of Kininogen in Human serum, plasma and cell culture media. An antibody specific for Human Kininogen was coated on a 96-well plate. Standards and samples are added to the wells and any Kininogen present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human Kininogen antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of Kininogen present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: Kininogen, also known as alpha 2-thiol proteinase inhibitor, is a multi-function protein. There are two alternatively spliced forms, designated as the high molecular weight (HMW) and low MW (LMW) forms . The HMW form is synthesized as a 644 amino acid (aa) precursor with a signal peptide. The mature chain is further processed into the heavy and the light chains. The active peptide bradykinin is released, which has a variety of functions including muscle contraction, hypotension and inflammation. The heavy chain consists of three cystatin-like domains, which are responsible for inhibiting cysteine proteases. The light chain consists of a His-rich domain, which is associated with the clotting activity. In comparison to the HMW form, the LMW Kininogen (427 aa) has the same sequence in its heavy chain and bradykinin, but a different sequence in its light chain. The LMW form is not involved in blood clotting.
