IL-7 is secreted constitutively into the conditioned medium of adherent bone marrowstromal cells and thymic cells. It stimulates the proliferation of pre-B and pro-B-cells withoutaffecting their differentiation and can replace murine bone marrow stromal cells insupporting the extended growth of both pre-B-cells and pro-B-cells. IL-7 may be of clinicalsignificance for Adoptive immunotherapy since it is capable in vivo to cause the CD4 (+) Tcell-dependent destruction of tumor cells.The Cell Sciences Human IL-7 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an invitro enzyme-linked immunosorbent assay for the quantitative measurement of human IL-7in serum, plasma, cell culture supernatants and urine. This assay employs an antibodyspecific for human IL-7 coated on a 96-well plate. Standards and samples are pipettedinto the wells and IL-7 present in a sample is bound to the wells by the immobilizedantibody. The wells are washed and biotinylated anti-human IL-7 antibody is added. Afterwashing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted tothe wells. The wells are again washed, a TMB substrate solution is added to the wells andcolor develops in proportion to the amount of IL-7 bound. The Stop Solution changes thecolor from blue to yellow, and the intensity of the color is measured at 450 nm.
