The Bioaim Human IL-29 ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of IL-29 in human serum and plasma. An antibody specific for human IL-29 was coated on a 96-well plate. Standards and samples are added to the wells and any IL-29 present binds to the immobilized antibody. The wells are washed and biotinylated anti-human IL-29 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of IL-29 present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: IL-29 is a member of the helical cytokine family and is a type III interferon. IL-29 plays an important role in host defenses against microbes and its gene is highly upregulated in cells infected with viruses. Human/mouse IL-28A (IFN-lambda 2), IL-28B (IFN-lambda 3), and human IL-29 (IFN-lambda 1) are class II cytokine receptor ligands that are distantly related to members of the IL-10 family. These cytokines exert bioactivities that overlap those of type I IFNs, including antiviral activity and up-regulation of MHC class I antigen expression. The proteins signal through the same heterodimeric receptor complex that is composed of the IL-10 receptor beta and a novel IL-28 receptor alpha.
