IL-15 (Interleukin-15) is found in conditioned medium of a monkey kidney epithelial cellline, CV-1/EBNA. It is a glycoprotein of 14-15 kDa. Human IL-15 has been cloned from ahuman stromal cell line, IMTLH. Stromal cell line IL-15 has been shown to be produced byhuman fetal astrocytes and microglia in response to IL-1a or IFN-a and may thus play arole in T-cell mediated immune responses in the human central nervous system. IL-15 is arecently identified novel cytokine that shares many biological properties with IL-2, includingT-Cell, B-Cell and NKcell-stimulatory activities.The Cell Sciences Human IL-15 ELISA (Enzyme-Linked Immunosorbent Assay) kit is anin vitro enzyme-linked immunosorbent assay for the quantitative measurement of humanIL-15 in serum, plasma (collect plasma using EDTA or heparin as an anticoagulant. Citrateplasma is not recommended for use in this assay), cell culture supernatants and urine.This assay employs an antibody specific for human IL-15 coated on a 96-well plate.Standards and samples are pipetted into the wells and IL-15 present in a sample is boundto the wells by the immobilized antibody. The wells are washed and biotinylated antihumanIL-15 antibody is added. After washing away unbound biotinylated antibody, HRPconjugatedstreptavidin is pipetted to the wells. The wells are again washed, a TMBsubstrate solution is added to the wells and color develops in proportion to the amountof IL-15 bound. The Stop Solution changes the color from blue to yellow, and the intensityof the color is measured at 450 nm.
