Interleukin-10 (IL10) is produced by activated CD8 (+) peripheral blood T-cells, by T-helper CD4 (+) T-cell clones after both antigen-specific and polyclonal activation, by B-cell lymphomas, and by monocytes following cell activation by bacterial LPS and mast cells. IL10 is a homodimeric protein with subunits having a length of 160 aa. IL10 inhibits the synthesis of many cytokines such as IL2 and TNF-beta in Th1 T-helper, but not of Th2 T-helper cells. IL10 also inhibits mitogen- or anti-CD3 induced proliferation of T-cells in the presence of accessory cells and reduces the production of IFN-gamma and IL2. It also down-regulates the expression of MHC class II Ags and co-stimulatory molecules on macrophages. It enhances B-cell survival, proliferation, and antibody production. IL10 can block NF-kappa B activity, and is involved in the regulation of the JAK-STAT signaling pathway. The Human IL10 ELISA is an, in vitro, enzyme-linked immunosorbent assay for the quantitative measurement of Human IL10 in serum, plasma, cell culture supernatants and urine. This assay employs an antibody specific for Human IL10 coated on a 96-well plate. Standards and samples are pipetted into the wells and IL10 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and Biotinylated Anti-Human IL10 antibody is added. After washing away unbound Biotinylated antibody, HRP-Streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IL10 bound.

Storage and Stability

May be stored for up to 6 months at 2-8C from the date of shipment. Standard should be stored below -20C (-80C recommended) after reconstitution. Opened Microplate Wells and reagents may be store for up to 1 month at 2-8C. Return unused wells to the pouch containing desiccant pack, reseal along entire edge of zipseal.
