The Bioaim Human GDNF ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of GDNF in human serum and plasma. An antibody specific for human GDNF was coated on a 96-well plate. Standards and samples are added to the wells and any GDNF present binds to the immobilized antibody. The wells are washed and biotinylated anti-human GDNF antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of GDNF present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: GDNF (Glial Cell Line-derived Neurotrophic Factor) is a secreted protein that promotes the survival of motoneurons, midbrain dopaminergic neurons, Purkinje cells, and sympathetic neurons. It is produced by Sertoli cells, type 1 astrocytes, Schwann cells, neurons, pinealocytes, and skeletal muscle cells. GDNF binding to GFR alpha 1 induces the recruitment of Ret, NCAM-1/CD56, various integrins, Syndecan-3, or N-Cadherin. GDNF-based therapies show promise in several neurodegenerative disorders, particularly Parkinson’s disease. GDNF also regulates kidney development and spermatogenesis, and it affects alcohol consumption.
