The Bioaim Human ErbB3 ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of ErbB3 in Human serum, plasma and cell culture media. An antibody specific for Human ErbB3 was coated on a 96-well plate. Standards and samples are added to the wells and any ErbB3 present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human ErbB3 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of ErbB3 present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.Research Area Cancer Biomarker;
erb-b2 receptor tyrosine kinase 3 (ErbB3) Background: ErbB3 has been shown to bind the ligands heregulin and NRG-2. Ligand binding causes a change in conformation that allows for dimerization, phosphorylation, and activation of signal transduction. ErbB3 can heterodimerize with any of the other three ErbB family members. The theoretical ErbB3 homodimer would be non-functional because the kinase-impaired protein requires transphosphorylation by its binding partner to be active. ErbB3 was found to be kinase impaired, having only 1/1000 the autophosphorylation activity of EGFR and no ability to phosphorylate other proteins.Therefore, ErbB3 must act as an allosteric activator. ErbB3 binding causes the allosteric activation of p110, the lipid kinase subunit of PI3K, a function not found in either EGFR or ErbB2.
