The Bioaim Mouse Adiponectin LISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of Adiponectin in mouse serum, plasma and cell culture media. An antibody specific for mouse Adiponectin was coated on a 96-well plate. Standards and samples are added to the wells and any Adiponectin present binds to the immobilized antibody. The wells are washed and biotinylated anti-mouse Adiponectin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of Adiponectin present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: Adiponectin (also known as Adipocyte complement-related protein of 30 kDa, Acrp30) is an adipocyte-derived protein with wide ranging paracrine and endocrine effects on metabolism and inflammation. It promotes adipocyte differentiation, fatty acid catabolism, and insulin sensitivity and is negatively correlated with obesity, type 2 diabetes, and atherogenesis. Adiponectin associates into 70 kDa noncovalently-linked trimers that are disulfide-linked into medium molecular weight hexamers and then into >300 kDa high molecular weight oligomers. Adiponectin is exclusively secreted from adipose tissue (and also from the placenta in pregnancy) into the bloodstream and is very abundant in plasma relative to many hormones. Levels of the hormone are inversely correlated with body fat percentage in adults; Similarly, circulating adiponectin concentrations increase during caloric restriction in animals and humans, such as in patients with anorexia nervosa.
