The Bioaim Human AgRP ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of AgRP in Human serum, plasma and cell culture media. An antibody specific for Human AgRP was coated on a 96-well plate. Standards and samples are added to the wells and any AgRP present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human AgRP antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of AgRP present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Agouti-related protein(AgRP)
Background: Agouti-related Protein (AgRP) is a neuropeptide and a functional antagonist of alpha-MSH on Melanocortin (MC)-3 and MC-4 receptors, both of which are implicated in obesity. Agouti-related protein is expressed primarily in the adrenal gland, subthalamic nucleus, and hypothalamus, with lower levels of expression in the testis, kidneys, and lungs. The appetite-stimulating effects of AgRP are inhibited by the hormone leptin and activated by the hormone ghrelin.  Ghrelin has receptors on NPY/AgRP neurons that stimulate the secretion of NPY and AgRP to increase appetite. AgRP is stored in intracellular secretory granules and is secreted via a regulated pathway. The transcriptional and secretory action of AgRP is regulated by inflammatory signals. It has been found that AgRP stimulates the hypothalamic-pituitary-adrenocortical axis to release ACTH, cortisol and prolactin.
