The Bioaim Rat Fas L ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of Fas L in rat serum, plasma and cell culture media. An antibody specific for rat Fas L was coated on a 96-well plate. Standards and samples are added to the wells and any Fas L present binds to the immobilized antibody. The wells are washed and biotinylated anti-rat FasL antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of Fas L present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.Research Area Apoptosis;

Fas ligand ( FasL ) Background: Fas Ligand (FasL), also known as CD178, CD95L, or TNFSF6, is a transmembrane protein homotrimer that binds to Fas/CD95 and triggers apoptosis in the Fas-expressing cell. Fas Ligand also binds the soluble decoy receptor DcR3. It is expressed on activated Th1 cells, CD8+ cytotoxic T cells, and NK cells. A soluble form can be released which remains trimeric and retains the ability to bind Fas. Fas Ligand-induced apoptosis plays a central role in the development of immune tolerance and the maintenance of immune privileged sites. Tumor cells can evade immune surveillance by up-regulating Fas Ligand to kill tumor infiltrating lymphocytes. In mice, a Fas Ligand point mutation is the cause of severe lymphoproliferation and systemic autoimmunity.
