The Bioaim Mouse TWEAK ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of TWEAK in mouse serum, plasma and cell culture media. An antibody specific for mouse TWEAK was coated on a 96-well plate. Standards and samples are added to the wells and any TWEAK present binds to the immobilized antibody. The wells are washed and biotinylated anti-mouse TWEAK antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of TWEAK present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.Research Area Apoptosis; Osteoclast differentiation;
tumor necrosis factor (ligand) superfamily, member 12 ( TWEAK ) Background: The protein encoded by this gene is a cytokine that belongs to the tumor necrosis factor (TNF) ligand family. This protein is a ligand for the FN14/TWEAKR receptor. This cytokine has overlapping signaling functions with TNF, but displays a much wider tissue distribution. Leukocytes are the main source of TWEAK including human resting and activated monocytes, dendritic cells and natural killer cells. TWEAK can induce apoptosis via multiple pathways of cell death in a cell type-specific manner. This cytokine is also found to promote proliferation and migration of endothelial cells, and thus acts as a regulator of angiogenesis.
