The Bioaim Mouse Thrombopoietin ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of Thrombopoietin (TPO) in mouse serum, plasma and cell culture media. An antibody specific for mouse Thrombopoietin was coated on a 96-well plate. Standards and samples are added to the wells and any TPO present binds to the immobilized antibody. The wells are washed and biotinylated anti-mouse TPO antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of TPO present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: Thrombopoietin (Tpo), is a key regulator of megakaryocytopoiesis and thrombopoiesis. It is principally produced in the liver and is bound and internalized by the receptor Tpo R/c-mpl. Defects in the Tpo-Tpo R signaling pathway are associated with a variety of platelet disorders. Tpo promotes the differentiation, proliferation, and maturation of MK and their progenitors. Tpo additionally promotes platelet production, aggregation, ECM adhesion, and activation. Aside from its hematopoietic effects, Tpo is expressed in the brain where it promotes the apoptosis of hypoxia-sensitized neurons and inhibits neuronal differentiation by blocking NGF induced signaling.
