The Bioaim Mouse Periostin ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of Periostin in mouse serum, plasma and cell culture media. An antibody specific for mouse Periostin was coated on a 96-well plate. Standards and samples are added to the wells and any Periostin present binds to the immobilized antibody. The wells are washed and biotinylated anti-mouse Periostin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of Periostin present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.Research Area Angiogenesis;
periostin, osteoblast specific factor (Periostin) Background: Periostin, also known as OSF-2, is a secreted matricellular protein with functions in extracellular matrix formation, cell migration, and inflammation. Periostin is expressed by mesenchymal cells such as vascular smooth muscle cells, fibroblasts, osteoblasts, and odontoblasts in developing teeth. It is upregulated in many carcinomas. Periostin binds to multiple Integrins and enhances cell adhesion and cell migration. It enhances Fibronectin and Collagen I production and promotes collagen fibrillogenesis. It also induces epithelial-mesenchymal transition, tumor growth, invasion, and metastasis. Periostin induces the expression of VEGF R2 on endothelial cells and VEGF-C in tumor cells, and it can induce tumor lymphangiogenesis. In many cancers, periostin binds to integrins on cancer cells, activating the Akt/PKB- and FAK-mediated signaling pathways. This leads to increased cell survival, invasion, angiogenesis, metastasis, and the epithelial-mesenchymal transition.
