The Bioaim Mouse MCP-5 ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of MCP-5 in mouse serum, plasma and cell culture media. An antibody specific for mouse MCP-5 was coated on a 96-well plate. Standards and samples are added to the wells and any MCP-5 present binds to the immobilized antibody. The wells are washed and biotinylated anti-mouse MCP-5 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of MCP-5 present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: Mouse MCP-5, also known as CCL12, encodes a 104 amino acid (aa) precursor protein with a 22 aa hydrophobic signal sequence that is cleaved to generate a 82 aa residue mature protein. MCP-5 is expressed constitutively in the thymus and lymph nodes. Under inflammatory conditions, MCP-5 expression is also induced in activated macrophages and mast cells. CCL12 specifically attracts eosinophils, monocytes and lymphocytes. It is thought to coordinate cell movements during early allergic reactions, and immune response to pathogens.
