The Bioaim Mouse M-CSF ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of M-CSF in mouse serum, plasma and cell culture media. An antibody specific for mouse M-CSF was coated on a 96-well plate. Standards and samples are added to the wells and any M-CSF present binds to the immobilized antibody. The wells are washed and biotinylated anti-mouse M-CSF antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of M-CSF present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: M-CSF, also known as CSF-1, is a four-alpha-helical-bundle cytokine that is the primary regulator of macrophage survival, proliferation and differentiation. M-CSF is also essential for the survival and proliferation of osteoclast progenitors. M-CSF also primes and enhances macrophage killing of tumor cells and microorganisms, regulates the release of cytokines and other inflammatory modulators from macrophages, and stimulates pinocytosis. M-CSF increases during pregnancy to support implantation and growth of the decidua and placenta. Sources of M-CSF include fibroblasts, activated macrophages, endometrial secretory epithelium, bone marrow stromal cells and activated endothelial cells. The M-CSF receptor (c-fms) transduces its pleotropic effects and mediates its endocytosis. Locally produced M-CSF in the vessel wall contributes to the development and progression of atherosclerosis. M-CSF has been described to play a role in renal pathology including acute kidney injury and chronic renal failure. The chronic activation of monocytes can lead to multiple metabolic, hematologic and immunologic abnormalities in patients with chronic renal failure. In the context of acute kidney injury, M-CSF has been implicated in promoting repair following injury, but also been described in an opposing role, driving proliferation of a pro-inflammatory macrophage phenotype.
