The Bioaim Mouse Endocan ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of Endocan in mouse serum, plasma and cell culture media. An antibody specific for mouse Endocan was coated on a 96-well plate. Standards and samples are added to the wells and any Endocan present binds to the immobilized antibody. The wells are washed and biotinylated anti-mouse Endocan antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of Endocan present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: Endocan, also known as Endothelial Cell-specific Molecule-1 (ESM-1) and IGFBP-rp6, is a secreted cysteine-rich dermatan sulfate proteoglycan. It is primarily expressed by endothelial cells within the vascular network of the kidney and in the alveolar walls of the lung. Endocan expression is upregulated by TNF-alpha, IL-1 beta and LPS, and downregulated by IFN-gamma. Endocan binds LFA-1 on human lymphocytes, monocytes and Jurkat cells, inhibiting its binding to ICAM-1 and reducing LFA-1-mediated leukocyte activation. Endocan expression has been shown to be increase in presence of pro-angiogenic growth factors such as VEGF (vascular endothelial growth factor) or FGF-2 (fibroblast growth factor 2). In hypervascularized cancers, overexpression of endocan has been detected by immunohistochemistry using monoclonal antibodies against endocan / ESM-1.
