The Bioaim Mouse E-Selectin ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of E-Selectin in mouse serum, plasma and cell culture media. An antibody specific for mouse E-Selectin was coated on a 96-well plate. Standards and samples are added to the wells and any E-Selectin present binds to the immobilized antibody. The wells are washed and biotinylated anti-mouse E-Selectin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of E-Selectin present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: The Selectin family is comprised of three members, E-Selectin, L-Selectin, and P-Selectin. E-Selectin is transiently expressed on vascular endothelial cells in response to IL-1 beta and TNF-alpha. L-Selectin is expressed constitutively on a wide variety of leukocytes. Two forms of L-Selectin have been reported, apparently arising as a result of post-translational modifications. Human P-Selectin (GMP-140, LECAM-3, PADGEM, CD62P) is expressed by activated platelets and endothelial cells. The extracellular domains of human and mouse P-Selectin share approximately 73% amino acid sequence identity.
