The Bioaim Mouse BLC ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of BLC in mouse serum, plasma and cell culture media. An antibody specific for mouse BLC was coated on a 96-well plate. Standards and samples are added to the wells and any BLC present binds to the immobilized antibody. The wells are washed and biotinylated anti-mouse BLC antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of BLC present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: CXCL13/BLC/BCA-1 is a constitutively expressed chemokine that plays an important role in B and T cell homing. It is expressed by salivary gland epithelium, dendritic cells, osteoclasts, and peritoneal macrophages. It can form homodimers or heterodimers with FGF basic, and it signals through CXCR3 or CXCR5. CXCL13 induces the migration of naïve B cells and a subset of memory T cells to lymphoid tissue. It also promotes B1 cell migration into the omentum and peritoneum. In the fetus, CXCL13 attracts CD4+ CD3- IL-7 R alpha+ hematopoietic cells to sites of future Peyer’s patch development.
