The Bioaim Mouse Amphiregulin ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of Amphiregulin in mouse serum, plasma and cell culture media. An antibody specific for mouse Amphiregulin was coated on a 96-well plate. Standards and samples are added to the wells and any Amphiregulin present binds to the immobilized antibody. The wells are washed and biotinylated anti-mouse Amphiregulin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of Amphiregulin present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: Amphiregulin is a member of the EGF family of cytokines, which is comprised of at least ten proteins including EGF, TGF-alpha, HB-EGF, and the various heregulins. All of these cytokines are synthesized as transmembrane precursors and are characterized by the presence of one or several EGF structural units in their extracellular domain. It is an autocrine growth factor as well as a mitogen for astrocytes, Schwann cells, fibroblasts. It is related to epidermal growth factor (EGF) and transforming growth factor alpha (TGF-alpha). This protein interacts with the Epidermal growth factor receptor (EGFR) to promote the growth of normal epithelial cells.
