The Bioaim Human WISP-1 ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of WISP-1 in Human serum, plasma and cell culture media. An antibody specific for Human WISP-1 was coated on a 96-well plate. Standards and samples are added to the wells and any WISP-1 present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human WISP-1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of WISP-1 present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.Research Area Angiogenesis; Aging;
WNT1 inducible signaling pathway protein 1(WISP-1) Background: WISP-1 belongs to the CCN (CTGF/CYR61/NOV) family of secreted proteins that share a common multimodular organization. Each family member has an IGFBP domain, a von Willebrand type C domain, a thrombospondin 1 domain and a C-terminal cysteine knot domain. WISP-1 promotes mesenchymal cell proliferation and osteoblastic differentiation, and represses chondrocytic differentiation. WISP-1 binds BMP2 and enhances BMP2 function in osteogenesis. These activities may be modulated by its direct binding to decorin and biglycan, two members of a family of small leucine-rich proteoglycans present in the extracellular matrix of connective tissue. WISP-1 attenuates p53-mediated apoptosis in response to DNA damage through activation of the Akt kinase, and inhibits TNF-induced cell death in cardiomyocytes.
