The Bioaim Human VCAM-1 ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of VCAM-1 in human serum and plasma. An antibody specific for human VCAM-1 was coated on a 96-well plate. Standards and samples are added to the wells and any VCAM-1 present binds to the immobilized antibody. The wells are washed and biotinylated anti-human VCAM-1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of VCAM-1 present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.Research Area Angiogenesis; Cancer Biomarker;cell adhesion, inflammation;
cullin 5 ( VACM-1 ) Background: VCAM-1 (Vascular Cell Adhesion Molecule-1; CD106) is a transmembrane molecule that mediates the adhesion of immune cells to the vascular endothelium during inflammation. It binds to several integrins including alpha4/beta1 (VLA-4), alpha4/beta7, alpha9/beta1, and alphaD/beta2. The VCAM-1 protein mediates the adhesion of lymphocytes, monocytes, eosinophils, and basophils to vascular endothelium. It also functions in leukocyte-endothelial cell signal transduction, and it may play a role in the development of atherosclerosis and rheumatoid arthritis. Upregulation of VCAM-1 in endothelial cells by cytokines occurs as a result of increased gene transcription (e.g., in response to Tumor necrosis factor-alpha (TNF-α) and Interleukin-1 (IL-1)) and through stabilization of Messenger RNA (mRNA) (e.g., Interleukin-4 (IL-4)). The promoter region of the VCAM-1 gene contains functional tandem NF-κB (nuclear factor-kappa B) sites. The sustained expression of VCAM-1 lasts over 24 hours.
