For the quantitative determination of Human TNF-alpha/TNFA/TNFSF2 concentration in cell culture supernates.
Human peripheral blood mononuclear cells ?2 x 10E6 cells/mL? were cultured in DMEM supplemented with 5% bovine calf serum, 50 µM ?-mercaptoethanol, 2 mM L-glutamine, 100 U/mL penicillin, and 100 µg/mL streptomycin sulfate, stimulated with 1 µg/mL PHA for 5 days. Aliquots of the cell culture supernates were removed and assayed for levels of natural TNFa, and measured 1144.57 pg/mL.
This assay recognizes both recombinant and natural Human TNFa. The factors listed below were prepared at 50 ng/mL in dilution buffer and assayed for cross-reactivity. No cross-reactivity was observed.
