The Bioaim Human Tie-2 ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of Tie-2 in Human serum, plasma and cell culture media. An antibody specific for Human Tie-2 was coated on a 96-well plate. Standards and samples are added to the wells and any Tie-2 present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human Tie-2 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of Tie-2 present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: Tie-2 is a receptor for the angiopoietin (ANG) family: ANG-1, ANG-2, and ANG-3 (mouse)/-4 (human). Ang-2 has been reported to act as an antagonist for Ang-1.This receptor possesses a unique extracellular domain containing 2 immunoglobulin-like loops separated by 3 epidermal growth factor-like repeats that are connected to 3 fibronectin type III-like repeats. Tie-2 is involved in vascular stabilization and remodeling.  Defects in TEK are associated with inherited venous malformations; the TEK signaling pathway appears to be critical for endothelial cell-smooth muscle cell communication in venous morphogenesis.
