The Bioaim Human TGF-beta RII ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of TGF-beta RII in Human serum, plasma and cell culture media. An antibody specific for Human TGF-beta RII was coated on a 96-well plate. Standards and samples are added to the wells and any TGF-beta RII present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human TGF-beta RII antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of TGF-beta RII present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: Most cell types express three sizes of receptors for TGF-beta. These are designated Type I, Type II and Type III. The Type I receptor is a membrane-bound serine/threonine kinase that apparently requires the presence of the Type II receptor to bind TGF-beta. The Type II receptor is also a membrane-bound serine/threonine kinase that binds TGF-beta1 and TGF-beta 3 with high affinity and TGF-beta 2 with much lower affinity.
