The Bioaim Human Prostasin ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of Prostasin in Human serum, plasma and cell culture media. An antibody specific for Human Prostasin was coated on a 96-well plate. Standards and samples are added to the wells and any Prostasin present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human Prostasin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of Prostasin present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: Prostasin, also known as channel activating protease 1, is a serine protease with trypsin-like substrate specificity. The preproenzyme possesses a C-terminal membrane-spanning domain that can be proteolytically processed to generate a secreted form of the enzyme. It is highly expressed in the prostate gland, and is also found at lower levels in the lung, kidney, liver, salivary gland, and pancreas. The peptidase activity of prostasin is involved in the regulation of epithelial sodium channels.
