The Bioaim Human PDGF-Rb ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of PDGF-Rb in human serum and plasma. An antibody specific for human PDGF-Rb was coated on a 96-well plate. Standards and samples are added to the wells and any PDGF-Rb present binds to the immobilized antibody. The wells are washed and biotinylated anti-human PDGF-Rb antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of PDGF-Rb present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: PDGF isoforms exert their cellular effects by binding to the structurally similar receptor tyrosine kinases (RTK), PDGF R alpha (CD140a) and PDGF R beta (CD140b). PDGF-AA, PDGF-BB, PDGF-CC, PDGF-DD and PDGF-AB isofoms bind the two distinct cell surface PDGF receptors with different affinities. Both PDGF R alpha and PDGF R beta are members of the class III subfamily of RTK which are characterized by the presence of five immunoglobulin-like domains in their extracellular region and a split kinase domain in their intracellular region. PDGF binding induces receptor homo- and heterodimerization leading to signal transduction. The expression of PDGF alpha and beta receptors is independently regulated in various cell types.
