The Bioaim Human P-Cadherin ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of P-Cadherin in human serum and plasma. An antibody specific for human P-Cadherin was coated on a 96-well plate. Standards and samples are added to the wells and any P-Cadherin present binds to the immobilized antibody. The wells are washed and biotinylated anti-human P-Cadherin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of P-Cadherin present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: Cadherin is a calcium-dependent cell-cell adhesion glycoprotein composed of five extracellular cadherin repeats, a transmembrane region and a highly conserved cytoplasmic tail. This gene is located in a six-cadherin cluster in a region on the long arm of chromosome 16 that is involved in loss of heterozygosity events in breast and prostate cancer. In addition, aberrant expression of this protein is observed in cervical adenocarcinomas. In addition to the classical cadherins, the cadherin superfamily also includes desmosomal cadherins, protocadherins, FAT family cadherins, and T-cadherin. In addition to cell adhesion, cadherins also mediate morphogenesis, cytoskeletal organization, and cell migration.
