The Bioaim Human OPN ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of OPN in human serum and plasma. An antibody specific for human OPN was coated on a 96-well plate. Standards and samples are added to the wells and any OPN present binds to the immobilized antibody. The wells are washed and biotinylated anti-human OPN antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of OPN present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.Research Area Angiogenesis; Cancer Biomarker;
secreted phosphoprotein 1 ( Osteopontin ) Background: Osteopontin (OPN) is also known as bone sialoprotein I (BSP-1 or BNSP), early T-lymphocyte activation (ETA-1), secreted phosphoprotein 1 (SPP1), 2ar and Rickettsia resistance (Ric). The central region of OPN contains RGD and non-RGD binding sites for multiple integrins. It is masked in full length OPN but is exposed following OPN cleavage by multiple proteases in tumors and sites of tissue injury. OPN is widely expressed and is prominent in mineralized tissues. It inhibits bone mineralization and kidney stone formation and promotes inflammation, cell adhesion, and migration.
