The Bioaim Human neprilysin ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of neprilysin in Human serum, plasma and cell culture media. An antibody specific for Human neprilysin was coated on a 96-well plate. Standards and samples are added to the wells and any neprilysin present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human neprilysin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of neprilysin present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.Research Area Angiogenesis;hematologic cancer;
membrane metalloendopeptidase (Neprilysin ) Background: Neprilysin is expressed at the cell surface of a variety of cell types. Enzymatically, neprilysin functions both as an endopeptidase with a thermolysin-like specificity and as a dipeptidylcarboxypeptidase. Neprilysin has been shown to be involved in the degradation of enkephalins in the mammalian brain and the inactivation of circulating atrial natriuretic peptide. Neprilysin has also been identified as the common acute lymphoblastic leukemia antigen (CALLA), and to be expressed on the surface of lymphocytes in some disease states. Neprilysin is also a major degrading enzyme of amyloid-beta peptide in the brain, indicating that down-regulation of neprilysin activity, which could be caused by aging, can contribute to the development of Alzheimers disease by promoting A-beta accumulation.
