The Bioaim Human MICB ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of MICB in Human serum, plasma and cell culture media. An antibody specific for Human MICB was coated on a 96-well plate. Standards and samples are added to the wells and any MICB present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human MICB antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of MICB present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: MICB is related to MHC class I and has similar domain structure, which is made up of external ⓫⓬⓭ domain, transmembrane segment and C-terminal cytoplasmic tail. MICB is a stress-induced ligand for NKG2D receptor. MHC class I chain-related genes MICA and MICB are transmembrane glycoproteins that function as ligands for human NKG2D. Recognition of MICA or MICB by NKG2D results in the activation of NK cell cytolytic activity and/or cytokine production. MICA/B are minimally expressed on normal cells, but are frequently expressed on or shed from epithelial tumors and can be induced by bacterial and viral infections.
