The Bioaim Human MCP-2 ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of MCP-2 in Human serum, plasma and cell culture media. An antibody specific for Human MCP-2 was coated on a 96-well plate. Standards and samples are added to the wells and any MCP-2 present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human MCP-2 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of MCP-2 present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: CCL8 is a small cytokine belonging to the CC chemokine family. The CCL8 protein is produced as a precursorcontaining 109 amino acids, which is cleaved to produce mature CCL8 containing 75 amino acids. The gene for CCL8 is encoded by 3 exons and is located within a large cluster of CC chemokines on chromosome 17q11.2 in humans. MCP-2 is chemotactic for and activates a many different immune cells, including mast cells,eosinophils and basophils, (that are implicated in allergic responses), and monocytes, T cells, and NK cellsthat are involved in the inflammatory response.
