The Bioaim Human Marapsin ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of Marapsin in Human serum, plasma and cell culture media. An antibody specific for Human Marapsin was coated on a 96-well plate. Standards and samples are added to the wells and any Marapsin present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human Marapsin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of Marapsin present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: Marapsin, pancreasin, and channel-activating protease 2 (CAP-2) are different names for the same serine protease that is expressed strongly in the pancreas. Synthesized as a single-chain proenzyme, marapsin is activated to form a disulfide bond-linked dimer between the pro peptide and the serine protease domain.
