The Bioaim Human LIMP-II ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of LIMP-II in Human serum, plasma and cell culture media. An antibody specific for Human LIMP-II was coated on a 96-well plate. Standards and samples are added to the wells and any LIMP-II present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human LIMP-II antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of LIMP-II present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: LIMPII (lysosomal integral membrane protein II), also known as LPG85 (85 kDa lysosomal membrane sialoglycoprotein) and CD36 antigen- like 2 , is a major lysosomal membrane protein. It belongs to the scavenger receptor class B subfamily and is designated member 2 (SR-B2).  LIMP2 has recently been identified as a novel component of intercalated discs in cardiac muscle. Intercalated discs are composed of gap junctions, adherens junctions and desmosomes, and are critical for the mechanical and electrical coupling of adjacent cardiomyocytes.
