The Bioaim Human Leptin ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of Leptin in human serum and plasma. An antibody specific for human Leptin was coated on a 96-well plate. Standards and samples are added to the wells and any Leptin present binds to the immobilized antibody. The wells are washed and biotinylated anti-human Leptin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of Leptin present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: Leptin is a hormone made by adipose cells that helps to regulate energy balance by inhibiting hunger. It is opposed by the actions of the hormone ghrelin, the “hunger hormone”. Both hormones act on receptors in the arcuate nucleus of the hypothalamus to regulate appetite to achieve energy homeostasis. In obesity, a decreased sensitivity occurs, resulting in an inability to detect satiety despite high energy stores. Although regulation of fat stores is deemed to be the primary function, it also plays a role in other physiological processes, as evidenced by its multiple sites of synthesis other than fat cells, and the multiple cell types beside hypothalamic cells that have its receptors. Many of these additional functions are yet to be defined.
