The Bioaim Human LDL R ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of LDL R in Human serum, plasma and cell culture media. An antibody specific for Human LDL R was coated on a 96-well plate. Standards and samples are added to the wells and any LDL R present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human LDL R antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of LDL R present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.Research Area Lipid metabolism;
low density lipoprotein receptor (LDL R) Background: LDL R is a widely expressed cell surface scavenger receptor. LDL R binds ApoB and ApoE, the apolipoproteins of low- and very low-density lipoproteins, respectively. It is a cell-surface receptor that recognizes the apoprotein B100, which is embedded in the outer phospholipid layer of LDL particles. The receptor also recognizes the apoE protein found in chylomicron remnants and VLDL remnants (IDL). It belongs to the Low density lipoprotein receptor gene family. It is most significantly expressed in bronchial epithelial cells and adrenal gland and cortex tissue. Hepatocyte LDL R is responsible for endocytosis and clearing of most plasma LDL cholesterol. At the low pH of the endocytic vesicle, it dissociates, allowing degradation of LDL and recycling of LDL R to the cell surface. Lack of LDL R expression or function causes familial hypercholesterolemia.
