The Bioaim Human IL-3 ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of IL-3 in human serum and plasma. An antibody specific for human IL-3 was coated on a 96-well plate. Standards and samples are added to the wells and any IL-3 present binds to the immobilized antibody. The wells are washed and biotinylated anti-human IL-3 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of IL-3 present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.

Background: Interleukin-3 (IL-3) is a secreted cytokine that stimulates the proliferation and differentiation of pluripotent hematopoietic stem cells and various lineage-committed progenitors. It also regulates the activation of mature mast cells, basophils, eosinophils, and macrophages. It is produced by activated T cells, thymic epithelial cells, activated mast cells, keratinocytes, neurons, and astrocytes. IL-3 signals through a receptor complex that contains IL-3 R alpha/CD123 and the common beta chain which is also a component of the receptors for IL-5 and GM-CSF. Receptors for IL-3 are present on bone marrow progenitors, macrophages, mast cells, eosinophils, megakaryocytes, basophils, and various myeloid leukemic cells.
