The Bioaim Human IL-2 R alpha ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of IL-2 R alpha in Human serum, plasma and cell culture media. An antibody specific for Human IL-2 R alpha was coated on a 96-well plate. Standards and samples are added to the wells and any IL-2 R alpha present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human IL-2 R alpha antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of IL-2 R alpha present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.Research Area Cancer Biomarker;
interleukin 2 receptor subunit alpha (IL-2 R α) Background: CD25/IL-2 receptor alpha is a transmembrane protein that associates with IL-2 R beta/CD122 and the Common gamma Chain/CD132 on activated T cells and regulatory T cells (Treg). The Common gamma chain is a shared subunit in the receptors for IL-4, -7, -9, -15, and -21. IL-2 R alpha is required for mediating IL-2 induced effects including T cell proliferation, activation induced cell death (AICD) of autoreactive naive T cells, and the development of CD4+CD25+ Treg which contribute to peripheral T cell homeostasis. A soluble form of IL-2 R alpha can be generated by proteolytic cleavage of the cell surface receptor. Infection by the protozoan Trypanosoma cruzi causes Chagas disease, characterized by a reduction in the amount of IL2RA expressed on the surface of immune cells. This leads to chronic immune suppression, becoming increasingly severe over the course of many years and ultimately resulting in death if left untreated.
