Interleukin-2 (IL2) is a secreted cytokine that is important for the proliferation of T- and B-lymphocytes. It is produced mainly by T-cells expressing the surface antigen CD4 following cell activation by mitogens or allogens under physiological conditions. IL2 displays significant anti-tumor activity for a variety of tumor cell types since it supports the proliferation and clonal expansion of T-cells that specifically attack certain tumor types. The receptor of this cytokine is a heterotrimeric protein complex whose gamma chain is also shared by Interleukin-4 (IL4) and Interleukin-7 (IL7). The targeted disruption of a similar gene in mice leads to ulcerative colitis-like disease, which suggests an essential role of this gene in the immune response to antigenic stimuli. The Human IL2 ELISA is an, in vitro, enzyme-linked immunosorbent assay for the quantitative measurement of Human IL2 in serum, plasma, cell culture supernatants and urine. This assay employs an antibody specific for Human IL2 coated on a 96-well plate. Standards and samples are pipetted into the wells and IL2 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and Biotinylated Anti-Human IL2 antibody is added. After washing away unbound Biotinylated antibody, HRP-Streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IL2 bound.
Storage and Stability
May be stored for up to 6 months at 2-8C from the date of shipment. Standard should be stored below -20C (-80C recommended) after reconstitution. Opened Microplate Wells and reagents may be store for up to 1 month at 2-4C. Return unused wells to the pouch containing desiccant pack, reseal along entire edge of zipseal.
