The Bioaim Human IGF-II ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of IGF-II in human serum and plasma. An antibody specific for human IGF-II was coated on a 96-well plate. Standards and samples are added to the wells and any IGF-II present binds to the immobilized antibody. The wells are washed and biotinylated anti-human IGF-II antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of IGF-II present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.Research Area cancer Biomarker; Aging;
insulin-like growth factor 1(IGF-1 ) Background: Insulin-like growth factor I and insulin-like growth factor II belong to the family of insulin-like growth factors that are structurally homologous to proinsulin. Both IGF-I and IGF-II are expressed in many tissues and cell types and may have autocrine, paracrine and endocrine functions. IGF-II is a potent mitogenic growth factor. However, unlike IGF-I which has important postnatal roles, the growth-promoting function of IGF-II is limited to embryonic development. Two specific cell surface receptors that bind IGF-I and IGF-II have been identified. The type I IGF receptor that participates in IGF signaling is structurally related to the insulin receptor. Type I IGF receptor binds IGF-I with higher affinity than IGF-II. The type II IGF receptor which binds IGF-II with much higher affinity than IGF-I is also the cation-independent mannose 6-phosphate receptor. Circulating IGFs exist in complexes bound to IGF binding proteins.
