The Bioaim Human HGF R ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of HGF R in Human serum, plasma and cell culture media. An antibody specific for Human HGF R was coated on a 96-well plate. Standards and samples are added to the wells and any HGF R present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human HGF R antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of HGF R present in the initial sample. MET proto-oncogene, receptor tyrosine kinase(HGFR) Background: HGF receptor, a product of the proto-oncogene c-met, is a heterodimeric transmembrane glycoprotein that is a receptor-type tyrosine kinase. c-MET is synthesized as a single-chain precursor (pr170) which undergoes post-translational glycosylation and proteolytic cleavage to give rise to the heterodimeric mature form. MET activation by its ligand HGF induces MET kinase catalytic activity, which triggers transphosphorylation of the tyrosines Tyr 1234 and Tyr 1235. These two tyrosines engage various signal transducers, thus initiating a whole spectrum of biological activities driven by MET, collectively known as the invasive growth program. The transducers interact with the intracellular multisubstrate docking site of MET either directly, such as GRB2, SHC, SRC, and the p85 regulatory subunit of phosphatidylinositol-3 kinase (PI3K), or indirectly through the scaffolding protein Gab1.
