The Bioaim Human HAI-2 ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of HAI-2 in Human serum, plasma and cell culture media. An antibody specific for Human HAI-2 was coated on a 96-well plate. Standards and samples are added to the wells and any HAI-2 present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human HAI-2 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of HAI-2 present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.Research Area Angiogenesis; cell adhesion. Apoptosis;
serine peptidase inhibitor, Kunitz type 2 (HAI-2) Background: Two alternatively spliced forms of HAI-2 are found in human tissues. HAI-2A, the full-length molecule and also known as placental bikunin, is a major form expressed in human tissues. HAI-2A consists of two Kunitz domains, and a C-terminal transmembrane domain. Both Kunitz domains can function as inhibitors independent of each other. In addition to HGF activator and trypsin, HAI-2A strongly inhibits plasmin, tissue and plasma kallikreins, and factor XIa. Three alternatively spliced forms of HAI-2 have been found in mouse tissues. HAI-2A is a minor form expressed in mouse tissues. HAI-2B, with residues 37 through 93 spliced out and thus lack of the first Kunitz domain, is a major form in mouse tissues. HAI-2B is an effective inhibitor of both HGF activator and trypsin.
