Description
Fas (APO-1 or CD95) is a cell-surface receptor that transduces apoptotic signals from Fas ligand (FasL). Fas and Fas Ligand(FasL) belong to the TNF superfamily and are type I and type II transmembrane proteins, respectively. Fas and FasL have beenobserved as soluble molecules in addition to their membrane associated forms. Fas is expressed to a large extent on activated Tand B lymphocytes, and on malignant lymphoid cells.The Cell Sciences Human Fas ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbentassay for the quantitative measurement of human Fas in serum, plasma, cell culture supernatants and urine. This assay employsan antibody specific for human Fas coated on a 96-well plate. Standards and samples are pipetted into the wells and Fas present ina sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-human Fas antibody isadded. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells areagain washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Fas bound. TheStop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
