The Bioaim Human Fas ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of Fas in Human serum, plasma and cell culture media. An antibody specific for Human Fas was coated on a 96-well plate. Standards and samples are added to the wells and any Fas present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human Fas antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of Fas present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.Research Area Apoptosis; metabolic regulation;
Fas cell surface death receptor (Fas) Background: Fas, also known as APO-1, CD95, and TNFRSF6, was originally identified as a cell-surface protein which binds to monoclonal antibodies that were cytolytic for various human cell lines. In the new TNF Receptor superfamily nomenclature, Fas is referred to as TNFRSF6. Fas is highly expressed in epithelial cells, hepatocytes, activated mature lymphocytes, virus-transformed lymphocytes and other tumor cells. Fas expression has also been detected in mouse thymus, liver, heart, lung, kidney and ovary. The ligand for Fas (FasL) has been identified and shown to be a member of the TNF family of type 2 membrane proteins.
