The Bioaim Human FAP ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of FAP in Human serum, plasma and cell culture media. An antibody specific for Human FAP was coated on a 96-well plate. Standards and samples are added to the wells and any FAP present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human FAP antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of FAP present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.Research Area Cancer Biomarker;
fibroblast activation protein alpha(FAP) Background: FAP is a type II transmembrane serine protease that degrades gelatin, Type I collagen, and N-terminal prolines. It is selectively expressed in reactive stromal fibroblasts of epithelial cancers, granulation tissue of healing wounds, and malignant cells of bone and soft tissue sarcomas. This protein is thought to be involved in the control of fibroblast growth or epithelial-mesenchymal interactions during development, tissue repair, and epithelial carcinogenesis. FAP expression is seen on activated stromal fibroblasts of more than 90% of all human carcinomas. Stromal fibroblasts play an important role in the development, growth and metastasis of carcinomas.
