The Bioaim Human EDA-A2 ELISA kit is a solid phase sandwich ELISA (enzyme-linked immunosorbent assay) for the quantitative measurement of EDA-A2 in Human serum, plasma and cell culture media. An antibody specific for Human EDA-A2 was coated on a 96-well plate. Standards and samples are added to the wells and any EDA-A2 present binds to the immobilized antibody. The wells are washed and biotinylated anti-Human EDA-A2 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is added to the wells. The wells are again washed and TMB substrate solution is added, which produces a blue color in direct proportion to the amount of EDA-A2 present in the initial sample. The Stop Solution changes the color from blue to yellow, and the microwell absorbances are read at 450 nm.Research Area Osteoclast differentiation;
ectodysplasin A (EDA-A2) Background: The products of the Ectodysplasin A gene (EDA, also called Tabby) are type II transmembrane proteins that belong to the TNF superfamily. The EDA gene encodes two splice variants, designated EDA-A1 and EDA-A2, that differ only by the deletion of two amino acids in the extracellular domain of EDA-A2. Despite this minor difference, the EDA isoforms display strong receptor specificity. EDA-A1 only binds EDAR, a member of the TNF Receptor superfamily (TNFRSF), whereas EDA-A2 binds to XEDAR, an X-linked TNFRSF member with high homology to EDAR. Along with c-Met, it has been shown to be involved in the differentiation of anatomical placodes, precursors of scales, feathers and hair follicles in vertebrates. Defects in this gene are a cause of ectodermal dysplasia, anhidrotic, which is also known as X-linked hypohidrotic ectodermal dysplasia.
